发酵虫草菌粉生产过程多糖类近红外光谱预测模型与质量控制

摘要

目的 采用近红外光谱进行发酵虫草菌粉生产过程中总多糖的快速测定,实现生产过程的含量测定和质量控制。方法 采用苯酚-硫酸法测定不同批次生产过程链菌种培养、菌种传代、菌种发酵、发酵物滤过、发酵物干燥、发酵物粉碎、发酵物混合各个操作单元中的总多糖含量,结合近红外光谱建立发酵虫草菌粉生产过程链总多糖偏最小二乘回归法(PLSR)定量分析模型,分析发酵虫草菌粉生产过程链中不同操作单元的质控水平。结果 生产过程链中发酵菌粉总多糖含量比较稳定;总多糖模型校正集均方差(RMSEC)、验证集均方差(RMSEP)、校正集相关系数(Rc)及验证集相关系数(Rp)分别为0.018 6%、0.013 3%、0.941 8和0.980 0;模型预测值与苯酚-硫酸法测定值之间误差率较小、拟合度较好。结论 该模型能实现快速分析发酵虫草菌粉质量,为生产过程链发酵虫草菌粉总多糖质控水平提供借鉴。

Abstract

OBJECTIVE To determine the total polysaccharides in the production processof fermented Cordyceps powder roduction, realize the content determination and quality control of theproduction process using near infrared pectroscopy. METHODS The phenol-sulfuric acid method was used to measure the total polysaccharides content in 30 batches various operating units of production process chain, such as strain culture, strain passage, strain fermentation, ferment filtration, ferment drying, ferment pulverizing, and ferment mixing,and combined with near infrared spectroscopy (NIRS) to establish a total polysaccharides partial least squares regression (PLSR) quantitative analysis model for the production process chain of fermented Cordyceps powder,and to analyze the quality control level of different operating units in the production process chain of Fermentation Cordyceps powder. RESULTS The total polysaccharides content of the esvarious Fermented Cordyceps powder in the production process chain is relatively stable. The root mean square errors of calibration and prediction (RMSEC, RMSEP) and the correlation coefficients of calibration and prediction (Rc, Rp) of the total polysaccharides PLSR model were 0.018 6%, 0.013 3%, 0.941 8 and 0.980 0, respectively. The absolute errorbetween the predicted value and phenol-sulfuric acid method measured value is small of the model, the fitting degree is good. CONCLUSION It can quickly analyze the quality of fermented Cordyceps powder and provide a reference for the quality control of total polysaccharides in the production process chain fermentation Cordyceps powder.
